Xenon gas could protect against Alzheimer’s disease! The study found that Xenon gas inhalation suppressed neuroinflammation, reduced brain atrophy, and increased protective neuronal states in mouse models of Alzheimer’s disease. Results are published in Science Translational Medicine, and a phase 1 clinical trial of the treatment in healthy volunteers will begin in early 2025. “It is a very novel discovery showing that simply inhaling an inert gas can have such a profound neuroprotective effect,” said senior and co-corresponding author. “One of the main limitations in the field of Alzheimer’s disease research and treatment is that it is extremely difficult to design medications that can pass the blood-brain barrier—but Xenon gas does. Automated diagnosis of respiratory diseases from lung ultrasound videos ensuring XAI: an innovative hybrid model approach Our model TD-CNN-LSTM-LungNet attained a remarkable accuracy of 96.57% in classifying LUS videos into pneumonia, COVID-19, normal, and other lung disease classes, which is above compared to ten traditional transfer learning models experimented with in this study. The eleven-ablation case study reduced training costs and redundancy. K-fold cross-validation and accuracy-loss curves demonstrated model generalization. The DSS, incorporating Layer Class Activation Mapping and heatmaps, improved interpretability and reliability, and PDF graphs facilitated precise decision-making by identifying feature boundaries. Life expectancy by county and educational attainment in the USA, 2000–19: an observational analysis Nationally, in all years there was a clear educational gradient in life expectancy at age 25 years where those with higher levels of education had higher life expectancy: individuals who had completed a college degree had higher life expectancy compared with those who had completed some college education (by 0·3 to 2·0 years over the study period), those who had completed some college had higher life expectancy than the high-school graduate population (by 4·1 to 4·9 years over the study period), and those who had graduated high school had higher life expectancy than those with less than a high-school education (by 3·4 to 5·1 years over the study period). From 2000 to 2019, life expectancy increased by 2·5 years (95% uncertainty interval 2·4 to 2·6) for the college graduate population, 0·7 years (0·6 to 0·8) for the population who had completed some college, and 0·3 years (0·3 to 0·4) for the high-school graduate population; there was no net change for those with less than a high-school education (0·0 years [−0·1 to 0·1]). In every educational attainment population, life expectancy increased by more from 2000 to 2010, with diminishing gains or even declines from 2010 to 2019. Life expectancy varied at the county level; eg, in 2019, the IQR for the population with less than a high-school education was 67·4 to 72·1 years, whereas for the college graduate population it was 82·3 to 84·6 years among counties with unmasked estimates. The college graduate population had higher life expectancy than those who had completed some college in 1937 (81·1%) of 2389 counties (47·1% statistically significant), which in turn had higher life expectancy than the high-school graduate population in 2726 (97·7%) of 2791 counties (87·0% statistically significant), which in turn had higher life expectancy than the population with less than a high-school education in 2356 (95·9%) of 2456 counties (91·6%). Emotional support across adulthood: A 60-year study of men’s social networks. Past research shows that social networks get smaller with age. But not all types of relationships may shrink at the same rate or for similar reasons. In the present study, we used a unique data set from a sample of 235 men who were followed longitudinally for 71 years to examine how the general pattern of network shrinkage documented in previous research generalizes to the number of emotional support providers in people’s networks. We additionally examined early-life predictors of the size of later-life support network. Growth curve analyses revealed that, mirroring the more general pattern of network shrinkage, emotional support networks shrink by as much as 50% between the ages of 30 and 90, reflecting an average reduction from two to one support providers. Examining the associations between prospectively collected measures of childhood family environment and later-life emotional support, we also found that men who grew up in warmer family environments had larger support networks in adulthood. In contrast, childhood family socioeconomic status was not connected to the size of emotional support networks later in life. The generalizability of this work is limited by the use of an archival all-male sample from the United States. Despite this limitation, these findings make important contributions to our understanding of adult socioemotional development and underscore the importance of prospectively collected longitudinal data in developmental research. The future of cancer immunotherapy will come through mitochondria: Cancer cells hijack T cells A study reveals how cancer cells suppress immune responses by transferring mutated mitochondria to tumor-infiltrating T cells. Researchers identified mitochondrial DNA mutations in T cells that matched mutations in the cancer cells they infiltrated. This transfer impaired T cell metabolism and function, leading to senescence and a diminished ability to fight tumors. The transfer occurs via tunneling nanotubes and extracellular vesicles, with cancer-derived mitochondria resisting normal degradation processes. These dysfunctional mitochondria disrupt T cell energy production, leaving the immune cells unable to mount effective antitumor responses. The findings could explain why some patients fail to respond to immune checkpoint inhibitors like PD-1 blockers. Targeting this mitochondrial transfer mechanism might open new doors for the future of cancer immunotherapy. Association of muscle strength and cardiorespiratory fitness with all-cause and cancer-specific mortality in patients diagnosed with cancer: a systematic review with meta-analysis High muscle strength and CRF were significantly associated with a lower risk of all-cause mortality. In addition, increases in CRF were associated with a reduced risk of cancer-specific mortality. These fitness components were especially predictive in patients with advanced cancer stages as well as in lung and digestive cancers. This highlights the importance of assessing fitness measures for predicting mortality in cancer patients. New cell therapy raises hope for curing type 1 diabetes – "Never succeeded before" In a first-in-human study at the ATMP center at Uppsala University Hospital in Sweden, the world’s first transplantation of genetically modified, hypoimmune insulin-producing cells from the islets of Langerhans has been used as a treatment. The key aspect is that the transplantation was carried out without immunosuppression. The hope is that, in the long term, this patient group could be cured of type 1 diabetes. The American company Sana Biotechnology, which developed the hypoimmune technology, saw its stock soar after the first results were presented this week. The company’s shares rose by approximately 200 percent. For individuals with severe type 1 diabetes, a well-established treatment involving transplantation of islets of Langerhans, a type of cell cluster containing insulin-producing cells, has been available for over 20 years. This treatment involves isolating islets of Langerhans from a deceased donor's pancreas and transplanting them into a diabetes patient with the goal of achieving normal blood sugar control and independence from insulin. Associations between different coffee types, neurodegenerative diseases, and related mortality: findings from a large prospective cohort study Background Observational studies have suggested associations between amount of coffee consumption and decreased risk of neurodegenerative diseases. However, these studies do not consider differences among coffee types, including sweetened, unsweetened, caffeinated, and decaffeinated varieties. Objectives This study aims to identify associations between the consumption of various coffee types (sugar-sweetened, artificially sweetened, unsweetened, caffeinated, and decaffeinated) and risks of Alzheimer’s disease and related dementias (ADRD) and Parkinson’s disease (PD), along with related mortality. Methods This prospective study included 204,847 participants (44.7% males) from the UK Biobank. Cox proportional hazards models were used to assess the associations of coffee type with neurodegenerative outcome. On the basis of coffee consumption, participants were divided into 5 groups: non-coffee consumers, >0–1 cup/d, ≥1–2 cups/d, ≥2–3 cups/d, and ≥3 cups/d. Results Over a median follow-up of 9 y, the study documented 1696 cases of ADRD, 1093 cases of PD, and 419 neurodegenerative-related deaths. In the multivariate analysis, compared with non-coffee consumers, those with the highest intake of unsweetened and caffeinated coffee (≥3 cups/d) showed hazard ratios (95% confidence intervals) of 0.75 (0.62, 0.91) for ADRD, 0.71 (0.56, 0.91) for PD, and 0.67 (0.44, 1.01) for neurodegenerative-related death.
